I just dug up some new info on sanctuary 101, specifically about its purposes, real purposes.
In General, the objectives of this Asylum was the observation of the effects of being in a confined space for several generations and the monitoring of genetic changes occurring when crossing within a small group, within a few generations.
I found this info from a terminal in the citadel. In the game this infa why it closed, but in actual fact it is. The Wiki already did it.

I wouldn't say that the staff of the vault-Tek were really sick people, especially because it's all carried out with the support of the US government.
